Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1 to 8). Keratins demonstrate tissue and differentiation specific expression profiles. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Alpha-1 Antichymotrypsin (AACT) is a plasma protease inhibitor synthesized in the liver as a single glycopeptide chain. In human, the normal serum level of AACT is about one-tenth that of alpha-1-antitrypsin (AAT), with which it shares nucleic acid and protein sequence homology. Both are major acute phase reactants; their concentrations in plasma increase in response to trauma, surgery and infection. Elevated levels of AACT are widely, but not universally, reported in the cerebrospinal fluid and plasma of AD patients. Prostate-specific antigen (PSA) and its SDS-stable complex with AACT are in widespread use as markers for the diagnosis of prostate cancer. AACT deficiency may also be a possible cause of chronic liver disease. AACT antibody reacts with histiocytes and histiocytic neoplasms. This MAb recognizes an epitope within the 74-182 C-terminal sequence (11kD peptide fragment) of human serum Cellular Retinol Binding Protein 1 (CRBP 1), a single-chain glycoprotein belonging to the superfamily of hydrophobic molecule transporter proteins, which is responsible for transport of retinol (vitamin A1) from the liver to peripheral target tissues, like the eye, where it mediates the cellular uptake. CRBP 1 is synthesized by hepatic parenchymal cells where it becomes bound to its ligand retinol and is then released into the circulation, where it binds further to the protein transthyretin, to form a transporting complex, which is big enough not to be lost by filtration through the kidney glomeruli. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. This antibody recognizes a protein of 21 kDa, identified as the Bax protein. This MAb is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Bcl-2 blocks cell death following a variety of stimuli. Bax has extensive amino acid homology with Bcl-2 and it homodimerizes and forms heterodimers with Bcl-2.
